Column chromatography purifies bulk chemical mixtures. A glass column is packed with a Stationary Phase (polar silica gel). The mixture is loaded on top, and a liquid Mobile Phase (solvent) is continuously poured through.

Separation Mechanism:
Compounds that strongly interact with the polar silica (Highly Polar compounds) travel very slowly through the column. Non-polar compounds are poorly retained and are swept out (eluted) first. Changing the solvent polarity can dramatically speed up or slow down this process.
